Critical priority pathogens have globally disseminated beyond clinical settings threatening wildlife. Andean Condors are essentials for ecosystem health and functioning, but their populations are globally near threatened and declining due to anthropogenic activities. During a microbiological and genomic surveillance study of critical priority antibiotic-resistant pathogens we have identified pandemic lineages of multidrug-resistant extended-spectrum β-lactamase (ESBL)-producing Escherichia coli colonizing Andean Condors admitted at two wildlife rehabilitation centres in South America. Genomic analysis revealed presence of genes encoding resistance to hospital and health-care agents among international E. coli clones belonging to sequence types (STs) ST162, ST602, ST1196 and ST1485. In this regard, resistome included genes conferring resistance to clinically important cephalosporins [i.e., CTX-M-14, CTX-M-55 and CTX-M-65 extended-spectrum β-lactamase (ESBL) genes], heavy metals (arsenic, mercury, lead, cadmiumerved.The present work examines the relationships of objectively measured sedentary time and physical activity (PA) with maximal fat oxidation during exercise (MFO) and the intensity of exercise that elicits MFO (Fatmax ), in young and middle-aged men and women. A total of 121 young sedentary adults (81 women, age 22.1±2.12 years, body mass index 25.2±4.9 kg/m2 ) and 70 middle-aged sedentary adults (36 women, age 53.4±4.94 years, body mass index 26.8±3.8 kg/m2 ) participated in this cross-sectional study. PA was objectively assessed using a wrist-worn accelerometer for 7 consecutive days. Wrist accelerations were classified into sedentary time and PA of different intensity (light, moderate, vigorous, moderate-vigorous), taking into account age-specific cut-offs. MFO and Fatmax were determined for all subjects by indirect calorimetry, using a walking graded exercise test. No association was found between any of the sedentary time-related variables and MFO (all P≥0.05). However, several sedentary time-related variables were related to FATmax in young men and women (all P≤0.04). A positive relationship was also detected between PA and MFO in young and middle-aged women (P≤0.05), although this became non-significant after adjustment for cardiorespiratory fitness (P>0.05). The present results suggest that, depending on the characteristics of the study cohort, sedentary time and PA time may be related to MFO and FATmax during exercise. Future longitudinal and intervention studies are warranted to better understand the role of sedentary time and PA in metabolic flexibility during exercise. This article is protected by copyright. The effect of low donor BMI on recipient graft function is not clear. Scientific Registry of Transplant Recipients data on recipients of deceased-donor kidneys from 2000 to 2019 were categorized by donor BMI (donor BMI 27). Low BMI donors (LBD) were more likely to be younger, female, and white (all p less then 0.05). LBD were less likely to be Hispanic, diabetic, or have hypertension (all p less then 0.001). LBD recipients were more likely to be younger and female (both p less then 0.001). Low donor BMI was not significantly associated with recipient graft survival. Donor-recipient BMI difference did not correlate with an increased risk of graft failure. Similar results were obtained when donors were classified using body surface area (BSA). Small donor size in terms of BMI or BSA or a large discrepancy between donor and recipient size should not necessarily preclude transplantation of an otherwise acceptable kidney. However, a nano-LC/MS/MS-based assay can suffer from limited reproducibility and MS signal instability making it challenging to employ it as a robust analytical method for routine clinical applications. METHODS To tackle these limitations, parallel reaction monitoring (PRM)-based targeted protein quantification using normal-flow liquid chromatography coupled with high resolution, accurate mass instrumentation was evaluated as a possible alternative. Different from the conventional selected reaction monitoring (SRM) using triple quadrupole MS, the proposed strategy used high resolution orbitrap MS coupled with conventional normal-flow liquid chromatography.
