KGaA, Weinheim.BACKGROUND Platelet-rich plasma (PRP) is an emerging orthobiologic treatment for musculoskeletal conditions like osteoarthritis. Two studies have demonstrated the influence of longer duration exercise on PRP composition, but no study has ever explored the impact of high intensity interval exercise (HIIE) on PRP content. OBJECTIVE To quantify cellular and growth factor content changes in PRP after 4 min of HIIE. DESIGN Controlled laboratory pilot study. SETTING Academic sports medicine center. PARTICIPANTS Ten healthy volunteers (5 male, 5 female). INTERVENTION Volunteers had PRP prepared from 15 mL of whole blood using a single spin, plasma-based system (Autologous Conditioned Plasma [ACP]) immediately before and after 4-min of HIIE on a stationary exercise bike (Tabata protocol). MAIN OUTCOME MEASURE The PRP was sent for complete blood counts and enzyme-linked immunosorbent assay (ELISA) to quantify transforming growth factor (TGF)-β, platelet-derived growth factor (PDGF), insulin-like growth factor (IGF)-1 and vascular endothelial growth factor (VEGF). RESULTS Mean platelet count in PRP increased from 367.4 ± 57.5 k/uL to 497.7 ± 93.3 k/uL after 4 min of HIIE (P less then 0.001). Transforming growth factor (TGF)-β also increased from 8237.2 ± 7676.5 pg/mL to 21 535.7 ± 4062.6 pg/mL post-exercise (P = 0.004). The other cellular components (leukocytes, red blood cells and mean platelet volume) and growth factors (PDGF, IGF-1 and VEGF) were not significantly changed. CONCLUSIONS A short 4-min bout of HIIE significantly increased the total platelet count and TGF-β concentration in PRP. This article is protected by copyright. All rights reserved. All rights reserved.Choanoflagellates, unicellular eukaryotes that can form multicellular colonies by cell division and that share a common ancestor with animals, are used as a model system to study functional consequences of being unicellular versus colonial. This review examines performance differences between unicellular and multicellular choanoflagellates in swimming, feeding, and avoiding predation, to provide insights about possible selective advantages of being multicellular for the protozoan ancestors of animals. Each choanoflagellate cell propels water by beating a single flagellum and captures bacterial prey on a collar of microvilli around the flagellum. Formation of multicellular colonies does not improve the swimming performance, but the flux of prey-bearing water to the collars of some of the cells in colonies of certain configurations can be greater than for single cells. Colony geometry appears to affect whether cells in colonies catch more prey per cell per time than do unicellular choanoflagellates. Although multicellular choanoflagellates show chemokinetic behavior in response to oxygen, only the unicellular dispersal stage (fast swimmers without collars) use pH signals to aggregate in locations where bacterial prey might be abundant. Colonies produce larger hydrodynamic signals than do single cells, and raptorial protozoan predators capture colonies while ignoring single cells. In contrast, ciliate predators entrain both single cells and colonies in their feeding currents, but reject larger colonies, whereas passive heliozoan predators show no preference. There is a need to adequately prepare undergraduate nursing students for clinical placement in a mental health setting in relation to their skills and confidence. This study aimed to evaluate the effect of a mental health simulation workshop on the skills and confidence of nursing students in providing care to consumers living with a mental illness. The study also evaluated the design of the mental health simulation workshop from an educational perspective. A pre/post-test survey was administered to a cohort of N = 89 Australian pre-registration nursing students. Exploratory factor analysis identified three factors Mental health therapeutic engagement, mental health assessment skills, and mental health placement preparedness. Analyses of pre-post differences indicated that all three factors were significantly different between the initial and follow-up responses, with follow-up responses being more favourable. The findings of this study demonstrate that there is value in including mental health simulated patient exercise as part of the learning strategies in the curriculum of pre-registration nurses. This has implications for the quality of care in the clinical environment and level of preparedness of these students' nurses for mental health clinical placement where they will be providing care to consumers living with a mental illness under direct supervision. © 2020 Australian College of Mental Health Nurses Inc.Heart transplantation (HTx) is a treatment option for end-stage heart failure in children. HTx is limited by the availability and acceptability of donor hearts. Refusal of donor hearts has been reported to be common with reasons for refusal including preexisting donor characteristics. This review will focus on the impact of donor characteristics and comorbidities on outcomes following pediatric HTx. A literature review was performed to identify articles on donor characteristics and comorbidities and pediatric HTx outcomes. Weight-based matching is the most common form of matching in pediatric HTx with a donor-recipient weight ratio between 0.7 and 3 having limited impact on outcomes. From an age perspective, donors less then 50 years can be carefully considered, but the impact of ischemic time needs to be understood. To increase the donor pool, with minimal impact on outcomes, ABO-incompatible donors should be considered in patients that are eligible.Bulevirtide
